摘  要:目的探讨香菇多糖对肺结核大鼠炎症反应的影响,并初步探究其潜在的作用机制。方法通过尾静脉注射结核分枝杆菌构建肺结核大鼠模型。将造模成功的大鼠随机分为5组:模型组、利福平组、香菇多糖低剂量(25mg/kg)、中剂量(50mg/kg)、高剂量(100mg/kg)组,每组10只;另选取10只斯泼累格·多雷雄性大鼠作为对照组。药物治疗4周后,使用血液分析系统检测各组大鼠支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中的炎症细胞水平。苏木精一伊红(hematoxylin and eosin,HE)染色观察肺组织病理损伤情况。酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测肺组织中促炎因子[肿瘤坏死因子α(tumor necrosis factorα,TNF-α)、白细胞介素(interleukin,IL)-1β、IL-6]和抗炎因子IL-10水平。实时荧光定量聚合酶链式反应和蛋白质免疫印迹法检测转化生长因子β(transforming growth factor β,TGF-β)/果蝇母体对抗多潜能性基因同源物(drosophilamothers against decapentaplegic homolog,Smad)2信号通路相关蛋白信使核糖核酸(messenger ribonucleic acid,mRNA)和蛋白表达水平。结果与相比于对照组,模型组大鼠BALF中的炎症细胞水平明显增多(P<0.05),肺组织病理损伤和炎性细胞侵润明显增加,促炎因子TNF-α、IL-1β、IL-6水平明显升高(P均<0.05),IL-10水平明显降低(P<0.05),TGF-β/Smad2信号通路相关mRNA和蛋白表达水平明显升高(P<0.05)。相比于模型组,利福平组、香菇多糖低、中、高剂量组大鼠BALF中的炎症细胞水平明显降低(P均<0.05),肺组织病理损伤和炎性细胞侵润明显减轻,促炎因子TNF-α、IL-1β、IL-6水平明显降低(P均<0.05),IL-10水平明显升高(P<0.05),TGF-β、Smad2mRNA和磷酸化Smad2、TGF-β蛋白表达水平明显降低(P均<0.05),其中香菇多糖高剂量效果最佳,且各香菇多糖组间呈剂量依赖性关系。结论香菇多糖可减轻肺结核大鼠模型中的炎症反应,其作用机制�Objective The aim of this study was to investigate the impact of lentinan on the inflammatory response in rats with tuberculosis and to preliminary explore its underlying mechanism of action.Methods A tuberculosis rat model was established by tail vein injection of Mycobacterium tuberculosis.Successfully modeled rats were randomly divided into five groups:a model group,a rifampicin group,and low(25 mg/kg),medium(50 mg/kg),and high(100 mg/kg)lentinan dose groups,with 10 rats in each group.Additionally,10 SD male rats were selected as the control group.After 4 weeks of drug treatment,a blood analysis system was used to detect the number of inflammatory cells in the bronchoalveolar lavage fluid(BALF)of rats in each group.Histopathological damage to lung tissue was observed via hematoxylin and eosin(HE)staining.Enzyme-linked immunosorbent assay(ELISA)was used to detect levels of pro-inflammatory cytokines[tumor necrosis factorα(TNF-α),interleukin(IL)-1β,IL-6],and anti-inflammatory cytokine IL-10 in lung tissue.Quantitative real-time polymerase chain reactionq(qRT-PCR)and Western Blot were employed to detect mRNA and protein expression levels of proteins related to the transforming growth factor(TCF-β)/mothers against decapentaplegic homolog 2(Drosophila)(Smad2)signaling pathway.Results Compared with the control group,the model group exhibited a significant increase in the number of inflammatory cells in BALF(P<0.05),along with markedly enhanced pathological damage and inflammatory cell infiltration in lung tissue.Levels of pro-inflammatory cytokines TNF-α,IL-1β,and IL-6 were significantly elevated(P<0.05),while IL-10 levels were notably decreased(P<0.05).Additionally,messenger ribonucleic acid(mRNA)and protein expression levels of proteins related to the TGF-β/Smad2 signaling pathway were significantly increased(P<0.05).In comparison to the model group,the rifampicin group and the low,medium,and high lentinan dose groups showed a significant reduction in the number of inflammatory cells in BALF(P<0.05),with al
