摘  要:为研究脂肪酶催化合成乙酸乙酯的规律,探讨不同条件下脂肪酶催化合成乙酸乙酯的效能,采用单因素和正交试验方法,研究酶添加量、底物(乙醇和乙酸)浓度和反应温度对脂肪酶催化合成乙酸乙酯的影响。结果表明,随着酶添加量、底物浓度和反应温度的增加,乙酸乙酯的生成量增加;优化正交试验得到的酯化条件:酶添加量为0.5%,乙醇浓度为280 g/L,乙酸浓度为30 g/L,反应温度为35℃,在此条件下得到乙酸乙酯的生成量为(8.29±0.08) g/L,合成的动力学拟合效果较好,相关系数达到0.998 3。研究为脂肪酶在工业生物催化中的应用提供参考。To investigate the patterns of lipase-catalyzed synthesis of ethyl acetate and evaluate its efficiency under varied conditions,single-factor and orthogonal test methods were employed to study the effects of enzyme dosage,substrate concentrations(ethanol and acetic acid),and reaction temperature on the synthesis.Results indicated that ethyl acetate yield increased with the elevation of enzyme dosage,substrate concentrations,and temperature.Optimal esterification conditions derived from orthogonal tests were as follows:enzyme dosage 0.5%,ethanol concentration 280 g/L,acetic acid concentration 30 g/L,and reaction temperature of 35℃.Under these conditions,the ethyl acetate yield reached(8.29±0.08)g/L,good kinetic fitting performance(correlation coefficients is 0.9983).This study provides a reference for the industrial application of lipase in biocatalysis.
