摘  要:目的 对13株肯塔基沙门菌生化和血清型复核,并进行溯源、药敏检测、耐药及毒力基因分析,了解肯塔基沙门菌的传播方式、耐药及毒力情况,为防控提供依据。方法 使用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF MS)快速鉴定并按国标法进行复核以确认菌株型别;脉冲场凝胶电泳(PFGE)溯源分析菌株传播方式;药敏实验检测菌株耐药性;全基因组测序并分析,识别耐药基因和毒力岛、分析进化关系等。结果 MALDI-TOF MS快速鉴定到沙门菌;国标检测法确认这13株菌为肯塔基沙门菌,药敏结果显示所有菌株对3~14种抗生素耐药;PFGE分析到永川病例分离株和万州食品株相似度100%;全基因组测序分子血清分型为肯塔基沙门菌;基因组中存在7个毒力岛和多个耐药基因/位点和基因突变;统进化树显示病例菌株和万州食品分离菌株处于同一进化分支的末端。结论 MALDI-TOF MS鉴定结果早于国标法2 d;13株肯塔基沙门菌均为多重耐药菌株,携带多种耐药基因和7个毒力岛,提示菌株可能对多类抗生素/消毒剂耐药且有较强的致病能力;PFGE和WGS溯源分析永川病例株与万州食品株高度一致,提示食源性疾病传播方式。Objective To re-examine the biochemical and serotype characteristics of 13 strains of Salmonella Kentucky(S.Kentucky) in Chongqing,to understand the prevalence,transmission mode,resistance,and virulence of Salmonella Kentucky(S.Kentucky),and to provide a basis for the prevention and control of S.Kentucky disease.Methods The biochemical identification and serotyping of 13 S.Kentucky strains were conducted by using matrix assisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF MS),and the strain serotyping was confirmed according to the national standard method.Pulse field gel electrophoresis(PFGE) was performed to trace the strain transmission mode.Drug sensitivity test was conducted to detect bacterial resistance.The whole genome sequencing was performed on 13 strains,and the obtained sequencing data were further analyzed,including species identification,virulence and resistance analysis,and genomic evolution analysis.Results The 13 strains were quickly identified as Salmonella by MALDI-TOF MS.These strains were confirmed to be S.Kentucky by the national standard testing method.Drug sensitivity results showed that all strains were resistant to 3-14 antibiotics.PFGE analysis showed a 100% similarity between the bacterial strains isolated from Yongchuan cases and the strains isolated from food in Wanzhou.The whole genome sequencing data showed that the molecular serotyping was S.Kentucky.There were 7 virulence islands and multiple resistance genes/loci and gene mutations identified in the genome of the 13 strains.The phylogenetic tree showed that the strains of Yongchuan cases and the strains isolated from food in Wanzhou were clustered at the end of the same evolutionary branch.Conclusion MALDI-TOF can identify the strains to genera and the identification result could be 2 days earlier than the national standard method.All the 13 strains of K.Salmonella are multidrug-resistant strains,carrying multiple resistance genes and 7 virulence islands,indicating that the strains may be resistant to
