摘 要:构建力竭性运动小鼠模型,随机分为安静组、模型组、阳性药物组、奶浆菌多糖(LDP)低、高剂量组,采用行为学试验、生化指标检测和免疫印迹法(western blot)评估LDP的干预效果。结果表明:LDP可显著提高力竭性小鼠的体质量,延长旋转棒上的停留时间,改善在避暗法、Morris水迷宫中的表现,降低血清中乳酸(LD)、乳酸脱氢酶(LDH)、血尿素氮(BUN)含量,增加肝糖原(LG)和肌糖原(MG)储备,提高脑组织中的超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性,降低丙二醛(MDA)、活性氧(ROS)含量。LDP还可上调脑组织中内皮型一氧化氮合酶(eNOS)、谷氨酸(Glu)、γ-氨基丁酸(γ-GABA)含量,促进海马组织中脑源性神经营养因子(BDNF)、磷脂酰肌醇3-激酶(PI3K)、核因子红细胞系2相关因子2(Nrf2)和血红素加氧酶-1(HO-1)的表达。A mouse model of exhaustive exercise was established and randomly divided into sedentary control group, model group, positive drug group, and low and high dose Lactarius deliciosus Gray polysaccharide(LDP) group. Behavioral experiments, biochemical indicator detection, and western blot analysis were used to evaluate the interventional effects of LDP. The results showed that LDP significantly increased the body weight of the exhaustive mice, prolonged their stay on the rotating rod, improved their performance in the dark avoidance method and Morris water maze, reduced the levels of laticacid(LD), lactate dehydrogenase(LDH), and blood urea nitrogen(BUN) in serum, increased the reserves of liver glycogen(LG) and muscle glycogen(MG), enhanced the activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in brain tissue, and reduced the contents of malondialdehyde(MDA) and reactive oxygen species(ROS). LDP could also up-regulate the contents of endothelial nitric oxide synthase(eNOS), glutamic(Glu), and γ-aminobutyric acid(γ-GABA) in brain tissue, and promoted the expression of brain-derived neurotrophic factor(BDNF), phosphatidylinositol 3-kinase(PI3K), nuclear factor erythroid 2-related factor 2(Nrf2), and heme oxygenase-1(HO-1) in hippocampus tissue.
