摘  要:目的 探讨NO对遗传性癫痫易感大鼠 (P77PMC大鼠 )惊厥发作的影响。方法 选择惊厥评分 30~ 40的P77PMC大鼠 30只 ,分成 5组 ,侧脑室注射不同药物后 ,铃声刺激 ,观察、记录大鼠惊厥潜伏期、惊厥程度、持续时间等变化。结果 注射L NAME后大鼠惊厥评分从 32± 4 .5增加到 48± 2 .0 (P <0 .0 5) ,持续时间从 35± 4 .5s延长到 72± 8.5s(P <0 .0 1 ) ;L 精氨酸、硝普钠和MK 80 1使大鼠惊厥评分从 32± 4 .5分别降到 1 7± 6 .0、1 5± 5 .5和 1 3± 6 .5(P <0 .0 5~ 0 .0 1 ) ,持续时间从 33± 6 .6s分别缩短到 2 0± 6 .5s、1 8± 6 .5s和 1 5± 3 .5s(P <0 .0 5~ 0 .0 1 ) ,除MK 80 1使惊厥潜伏期从 45± 7.5s延长到 75± 8.5s(P <0 .0 1 )外 ,余对其无影响。结论 NO合成酶抑制剂能明显加重惊厥程度和延长持续时间 ,其合成底物或释放剂以及NMDA受体拮抗剂均能产生相反效果 ,提示NO可作为内源性抑制性调质 。Objective To investigate the effects of nitric oxide(NO)on convulsions in genetically epilepsy prone rat(P77PMC). Methods 30 rats with convulsion scores 30~40 were randomly divided into 5 groups respectively .The latency ,convulsion scores and duration of convulsions in P77PMC treated by various factors were observed. Results N nitro arginine methyl ester (L NAME) can increase convulsion scores from 32±4.5 to 48±2.0(P<0.05) and duration from 35±4.5 to 72±8.5 seconds(P<0.01), however,L arginine, SNP(donor of NO) and MK 801(NMDA receptor inhibitor )can decrease convulsion score from 34±6.5 to 17± 6.0 , 15 ±5.5, 13±6.5(P<0.01) and duration from 32 ±7.5 to 20±6.5, 18±6.5 and 15±3.5(P<0.05),and latent period were extended from 45±7.5 to 75±8.5 seconds only by MK 801(P<0.01). Conclusion Inhibitor of NOS can aggravate convulsion intensity, while substrate ,donor of NO and NMDA receptor inhibitor induced opposite effects. These results indicated that NO ,as a endogenous inhibitor, is involved in convulsion termination processes.
