摘 要:从实验室保藏的菌种中筛选到一株黑曲霉 (Aspergillusniger)SW - 33,能够将 1g L的阿魏酸底物转化为 0 .2 3g L的香草酸 ,相应的摩尔转化率为 2 9.35 % ;流加四次底物阿魏酸后 ,产物浓度达到 1.11g L ,相应的摩尔转化率为 4 4 .9%。为了提高产物浓度 ,对培养基和发酵条件进行优化 ,使得该菌株能够将 1g l的阿魏酸底物转化为 0 .4 6g L的香草酸 ,相应的摩尔转化率为 5 7.81%。提取得到的香草酸 ,经HPLC测定 ,纯度为 85 .9% ;提取收率为 75 .2 %。用含香草酸的转化液 ,或者用提取的结晶香草酸 ,加入朱红密孔菌 (Pycnporuscinnabarnus)SW - 0 2 0 3发酵培养液 ,可得到转化产物香草醛。Aspergillus niger SW-33 was isolated from the microorganisms.This strain could transform 1g/L ferulic acid to produce0.23g/L vanillic acid with a molar yield of 29.35%. After ferulic acid was sequentially added into the medium for four times, the concentration of vanillic acid reached 1.11g/L with a molar yield of 44.9%. In order to improve the yield of product, the optimum medium and fermentation conditions were selected. Under the optimum condition, 1g/L ferulic acid could be transformedby A.niger SW-33 into 0.46g/L vanillic acid with a molar yield of 57.81%. Vanillic acid was extracted from the mediumand measured by HPLC, the purity quotient was 85.9% , the recovery was 75.2%. When the medium containing vanillic acid or crystallize of vanillic acid was put into the medium containing Pycnporus cinnabarnus SW-0203,vanillic acid could be transformed into vanillin.
