摘  要:目的在肺癌基因疫苗对小鼠免疫原性及对荷瘤动物治疗作用研究基础上,探讨对肺癌患者的安全性和耐受性,初步观察疗效.方法γ-IFN基因利用逆转录病毒载体PLXSN导入人肺癌A549细胞中的疫苗,对13例肺癌患者皮下注射2×106~1×107细胞,每间隔7~10天一次,连续6次.结果3例患者肺部肿癌,纵隔转移的淋巴结明显缩小,2例骨转移疼痛缓解.CD4、MHC、NK、活化CD4升高.结论肺癌基因疫苗能使体内形成特异性细胞免疫应答,对肿瘤有抑制作用.Objective Based on the Study of the effect of transgenic human lung adenocarcinoma cell line A549/IFN -γ as tumor vaccine on immunogenicity and inhibition of animal tumor, to probe into Safety, tolerance and initial efficacy to the patients with lung adenocarcinoma . Methods Interferon - gamma (IFN - γ) gene was introduced into human lung adenocarcinoma cell line A549 and was expressed stably on the basis of the construction of retroviral packing cell line PA317/PLXSN, Total 2 ×106 - 1 × 107 cell was inoculated in subcutaneous of total 13 patients with lung cancer once every 7-10 day, total six times consecutively.Results The result indicated that lymphatic node of mediastinum was reduced obviously in three patients with lung cancer tumor pain of bone metastasis in two patients was lysed.CD4, MHC1, NK, activated CD4 were increased. Conclusion lung carcinoma gene vaccine is able to produce excellent immunol response of cell patients, which might be of suppression effect on lung tumor.
