高效液相色谱法测定棒状链霉菌发酵液中克拉维酸的含量

摘　　要：利用克拉维酸与咪唑试剂的衍生化反应 ,建立了用高效液相色谱仪测定发酵液中克拉维酸含量的方法。色谱条件为 :采用HypersilODS2柱 ( 5 μm ,4 6mm× 2 0 0mm ) ,流动相为 0 1mol/LKH2 PO4∶甲醇 ( 94∶6) ,流速 1 0ml/min ,检测波长 3 11nm。结果表明 ,克拉维酸量在 1～ 10 0 μg范围内 ,该方法的线性关系及测定结果的精密度、回收率均良好 ,是一种简便、快速、有效的方法。An HPLC method, utilizing the derivation of clavulanic acid with imidazole, was developed, by which the content of clavulanic acid in fermentation broth was quantitatively measured. HPLC conditions were as follows, column: reverse phase C 18 , Hypersil ODS2 (5μm, 4 6mm×200mm); mobile phase: 0.1 mol/L KH 2PO 4 -methanol (94∶6); flow rate: 1.0ml/min; detection wavelength: UV 311nm. Results showed that the measurement was precise and linear. The recovey rate was high and this method was simple, rapid and reliable.
