摘 要:目的 探讨细胞周期调控抑制因子p5 7kip2 蛋白和cyclinE(周期蛋白E)在大肠腺癌发生发展中的作用。方法 采用免疫组化技术SP法 ,检测p5 7kip2 和cyclinE蛋白在 3 7例大肠腺癌和 2 6例正常大肠粘膜组织中的表达。结果 p5 7kip2 蛋白阳性表达率在大肠腺癌组织中为 40 5 % ,显著低于正常大肠粘膜组织 ( χ2 =5 0 3 9,P <0 0 5 ) ,并与大肠腺癌组织分化程度和淋巴结转移均有关 ( χ2 =8 914 ,χ2 =4 416,P <0 0 5 ) ;cyclinE蛋白阳性表达率在大肠腺癌组织中为 64 9% ,显著高于正常大肠粘膜组织 ( χ2 =4 2 85 ,P <0 0 5 ) ,并与大肠腺癌组织分化程度和淋巴结转移均有关 ( χ2 =8 3 97,χ2 =5 2 61,P <0 0 5 ) ;p5 7kip2 蛋白阳性表达组cyclinE蛋白阳性表达率低于p5 7kip2 蛋白阴性表达组 ,但两者之间呈负相关 ( r=-0 3 0 2 7,P <0 0 5 )。结论 p5 7kip2 蛋白的低表达和 (或 )cyclinE蛋白的高表达可能在大肠腺癌发生发展中发挥重要作用 ;p5 7kip2Objectives To investigate the effects of inhibiting factor of cell cycle regulation- p57 kip2 and cyclin E proteins in the genesis and progression of human colorectal adenocarcinoma.Methods The expressions of p57 kip2 and cyclin E proteins in tumor tissues of 37 patients with colorectal adenocarcinoma and in normal colorectal mucosal tissues of 26 subjects were detected by SP immunohistochemical technique.Results p57 kip2 protein positive-expression rate in tumor tissues of colorectal adenocarcinoma was 40 5%,which was lower than that in normal colorectal mucosal tissues (χ 2=5 039,P<0 05),p57 kip2 protein positive-expression correlated significantly with tumor cell differentiation and lymph node metastasis (χ 2=8 914;χ 2=4 416,P<0 05);cyclin E positive-expression rate in tumor tissues was 64 9%,which was higher than that in normal colorectal mucosal tissues (χ 2=4 285,P<0 05);cyclin E positive-expression also correlated significantly with tumor cell differentiation and lymph node metastasis (χ 2=8 397;χ 2=5 261,P<0 05);cyclin E protein positive-expression rate in the tumor tissues of p57 kip2 protein positive-expression group was lower than that in p57 kip2 protein negative-expression group,there were significant negative correlation between the two groups (r=-0 3027,P<0 05).Conclusions The decreased expression of p57 kip2 protein and/or over-expression of cyclin E protein may play an important role in the genesis and progression of colorectal adenocarcinoma.There is negative feedback mechanism between p57 kip2 and cyclin E proteins in cell cycle regulation of colorectal adenocarcinoma.
