绿茶对乳腺癌细胞血管内皮生长因子表达的影响及机制

摘　　要：目的　研究绿茶对人乳腺癌细胞株血管内皮生长因子 (VEGF)表达的效应及其机制。方法　VEGF蛋白的测定采用酶联免疫吸附法 ;Northern印迹杂交法检测mRNA表达 ,含VEGF启动子的报告基因来检测VEGF转录活性。结果　绿茶有效成分在 48h时分别抑制89 .71%乳腺癌细胞细胞分泌的及 3 3 .93 %细胞内合成的VEGF蛋白 ,48h抑制 5 5 .5 7%的VEGF转录活性 ,在 2 4、48h绿茶成分可明显抑制VEGF的mRNA表达 ,激活蛋白 1(AP 1)的抑制剂抑制了VEGF的mRNA表达 ,绿茶有效成分可进一步增强这种抑制作用。结论　绿茶能够抑制乳腺癌细胞株VEGF水平的表达 ,这种抑制可能通过包括AP 1在内的多途径实现。Objective To study the role of green tea on vascular endothelial growth factor (VEGF) in cancer cells and its mechanism.Mehtods Protein concentrations were determined by ELISA.Northern blot was performed to detect the expression of mRNA and promoter activity was analyzed by report gene technology.Results Green tea active components decreased 89.71% of secreted VEGF protein as well as 33.93% of synthesized VEGF protein in breast cancer cells after treatment for 48 h.Compared with control the components decreased 55.57% VEGF promoter activity in 48 h and also the VEGF mRNA in breast cancer cells in 24 h and 48 h.Furthermore the suppression of VEGF by AP-1 activity inhibitor was enhanced by green tea.Conclusion Green tea can inhibit VEGF expression in human breast cancer cells,which may be through multiply pathways besides AP-1.
