摘  要:提取扑草净降解优势菌 Pseudomonas FR粗酶液 ,以藻酸钠作为包埋剂将酶液固定化 ,比较固定化酶与游离酶对扑草净的作用效果 ,结果表明 ,理想条件下 ,固定化酶的活性相当于游离酶液活性的 79% ,但温度、p H值等条件的变化对固定化酶的影响很小 ;对固定化载体进行了优化 ,确定纤维系为适宜的添加剂 ,利用固定化酶对受污染土壤进行处理 ,经六周检测 ,土壤中扑草净的含量减少 85 % .The enzyme-extract of Pseudomonas FR, a super degrading-prometryn strain, has been studied in this paper. The degradative effect of immobilized enzyme-extract, embedded by sodium alginate, was compared with the effect of dissociated enzyme. The result showed that under the optimum condition, the activity of immobilized enzyme is about 79 per cent of dissociated enzyme's. However, as to immobilized enzyme, the effect of temperature and pH isvery low. Then the immobilized carriers were optimized, and cellulose was chosen as additive. The result of bioaugmentation with the immobilized enzyme showed the concentration of prometryn in soil was decreased by 85% after six weeks.
