摘  要:目的探讨苯妥英钠对脑缺血再灌注小鼠学习记功能保护作用的作用机制,为苯妥英钠用于脑缺血缺氧的治疗提供理论依据。方法采用清醒小鼠脑缺血再灌注手术方法,应用跳台法和避暗法,脑海马CA1区神经细胞计数等方法研究苯妥英钠对小鼠脑缺血再灌注行为学、脑病理形态学的影响。结果脑缺血再灌注组(模型组)小鼠错误次数明显增多:3.2±1.1(跳台)、8.4±3.2(避暗);逃避潜伏期(EL)延长:(37.5±15.0)s,出现明显的学习记忆功能障碍并使脑海马CA1区神经细胞数明显减少70±6神经细胞数(个/2mm2);苯妥英钠可较好地改善脑缺血再灌注致学习记忆障碍小鼠的学习记忆功能,错误次数减少:1.0±0.5(跳台),2.5±1.7(避暗);EL缩短:(6.7±1.9)s且使脑海马CA1区神经细胞数明显增加:76±6神经细胞数(个/2mm2)。结论苯妥英钠对脑缺血再灌注小鼠的学习记忆障碍有明显的改善作用,其作用机制可能与保护小鼠脑海马CA1区神经细胞有关。Aim To study the mechanism for Dephenylh ydantion(DPH)to improve learning-memory impairmen t of mice caused by cerebral Ischemi-a-reperfusion.Methods Forty Kunming mice were divided into four groups:control group,pseudo-operation group,model group and phenytoin group by using sortition method.The model an d phenytoin groups(experimental group)were treated with 40mg /kg DPH while p seudo-operation and control group(control group)treated with equivalent water.Step down test,step through test and the number of neurons of hippocam pus CA 1 were conducted to investigate the effect of DPH on the praxiology an d pathomorphology in mice with cereb ral ischemia-reperfusion.Results In comparison with the control group,the mice in the model group had obvious mistak es,including 3.2±1.1in step down test,8.4±3.2in step through test an d latent period prolonged(37.5±15.0)s,with the number of neurons of hippocampas CA 1 decreased(70.34±5.68)/2mm 2 while the learning-memory ability o f mice in the experimental group was greatly impr oved,including 1.0±0.5in step down test,2.5±1.7in step through test an d latent period shortened(6.7±1.9)swith the number of neurons of hippocampas CA 1 increased(76.28±6.47)/2mm 2 .Conclusion Dephenylhydantion improves the learning-memory im-pairment of mice caused by cerebral I schemia-reperfusion and its mechan ism may be related to protective neurons of hippocampas CA 1 .
