摘 要:以酶学分析的方法研究9种金属离子(Cu2+、Mg2+、Fe3+、Hg2+、Zn2+、Ca2+、Pb2+、Ag+和Mn2+)对缢蛏(Sinonovaculaconstricta)蛋白酶、淀粉酶和纤维素酶活力的影响。缢蛏壳长(5 64±0 25)cm,9种金属离子浓度分别设置为2×10-6mol/L、2×10-5mol/L、2×10-4mol/L、2×10-3mol/L、2×10-2mol/L5个梯度。实验表明,缢蛏3种消化酶活力大小为淀粉酶>蛋白酶>纤维素酶;在实验设置的浓度范围内,Cu2+、Fe3+、Mg2+、Pb2+对蛋白酶活力影响不显著,Hg2+、Zn2+、Ca2+、Ag+、Mn2+对蛋白酶活力影响显著;Mg2+、Pb2+对淀粉酶活力影响不显著,Cu2+、Fe3+、Hg2+、Zn2+、Ca2+、Ag+、Mn2+对淀粉酶活力影响显著;Ca2+、Mg2+、Zn2+对纤维素酶活力影响不显著,Pb2+、Cu2+、Fe3+、Hg2+、Ag+、Mn2+对纤维素酶活力影响显著。不同浓度金属离子对缢蛏蛋白酶、淀粉酶和纤维素酶活力的影响表现出不同的变化趋势。The concentrations of nine metal ions(Cu2+、Mg2+、Fe3+、Hg2+、Zn2+、Ca2+、Pb2+、Ag+ and Mn2+)were designed at 2×10-6 mol/L, 2×10-5 mol/L, 2×10-4 mol/L,2×10-3 mol/L and 2×10-2 mol/L, respectively, and the Sinonovacula constricta samples were employed with shell length at 4.25-6.85 cm. After 48 h starvation, the activities of protease, amylase and cellulase in the S. constricta were analyzed by using enzyme analytical method. The results indicate that the activity of amylase in S. constricta is the highest among the three digestive enzymes, and the activity of protease is higher than that of cellulase. Within the experimental concentration range, Cu2+,Fe3+,Mg2+ and Pb2+ have no significant effects, whereas Hg2+,Zn2+,Ca2+,Ag+ and Mn2+ have significant effects on protease activity of S. constricta; Mg2 and Pb2+ have no significant effects ,whereas Cu2+,Fe3+,Hg2+,Zn2+,Ca2+,Ag+ and Mn2+ have significant effects on amylase activity; Ca2+,Mg2+ and Zn2+ have no significant effects,whereas Pb2+,Cu2+,Fe3+,Hg2+,Ag+ and Mn2+ have significant effects on cellulase avtivity.The activities of protease, amylase and cellulase in S.constricta show different variation patterns under the different concentrations of metal ions.
