T淋巴瘤侵袭诱导基因在卵巢癌细胞株的表达及其意义

摘　　要：目的研究T淋巴瘤侵袭诱导基因1(Tiam1)在卵巢癌侵袭转移方面的作用。方法RT-PCR检测Tiam1和Rac1mRNA的表达;Westernblot法检测Tiam1蛋白质的表达;Boyden小室体外侵袭实验测定癌细胞迁移能力。结果Tiam1mRNA和蛋白质及Rac1mRNA在4株卵巢癌细胞A2780、Caov3、Skov3及SW626中均呈中高度表达;癌细胞平均迁移百分数分别为(27.67±3.2)%、(10±1.0)%、(22.67±2.5)%及(47.67±1.52)%。Tiam1及Rac1表达水平与癌细胞迁移潜能间均呈显著性相关,r分别为0.874和0.814,P=0.003和0.042。结论Tiam1/Rac1信号转导通路在卵巢癌细胞的侵袭和转移过程中起重要作用,为卵巢癌的基因治疗提供了新的靶点。To study the role of T lymphoma invasion/metastasis gene1(Tiam1)and protein in ovarian tumor cells.Methods Expressions of Tiam1mRNA,Rac1mRNA,and Tiam1protein in four ovarian tumor cells A2780,Caov3,Skov3,and SW626were studied by using RT-PCR and Western blot,respectively.The cell migration ability was analyzed by in vitro invasion assay.Results Expressions of Tiam1mRNA and protein,as well as Rac1mRNA were detected in all four ovarian tumor cells.There was a strong direct correlation between the levels of Tiam1and Rac1mRNA expression and migration potentials of all four ovarian cancer cells in vitro experiments.The increased expressions of Tiam1mRNA were coincident with those of Rac1mRNA,with a parallel relationship(P=0.003,r=0.874).Levels of Rac1mRNA expression were significantly correlated with the potentials of tumor cell migration(P=0.042,r=0.814).Conclusion Tiam1-Rac1signaling pathway plays a positive role in assessing tumor cell invasion and metastasis and provides a new target for gene therapy of ovarian cancer.
