摘 要:目的 探讨拓扑替康 (topotecan ,TPT)对肝癌细胞增殖和凋亡的生物学效应及机制。方法 采用HE染色、MTT实验、克隆形成试验、流式细胞仪、DNA电泳、透射电镜观察等方法 ,对TPT在HepG2细胞株的作用进行体外研究。结果 TPT对HepG2细胞产生明显的生长抑制作用 ,并可诱导HepG2细胞凋亡 ,与细胞周期被阻滞于S期有关。 结论 TPT对肝癌细胞具有显著的体外生长抑制及凋亡诱导作用 ,提示TPT是治疗肝癌潜在的分子药物。Objective To investigate the biological effects of topotecan(TPT) and its mechanism on the proliferation and apoptosis of human hepatocarcinoma cell line.Methods The effects of TPT of HepG2 cell line were investigated in vitro with HE staining,MTT,clong froming,flow cytometry,DNA gel electrophoresis and transmission electron microscope.Results TPT had marked suppressive effect on the growth of HepG2 cell line,and TPT could induce the apoptosis of HepG2 with the cell cycle arrested on S phase.Conclusion TPT can inhibit the growth and induce the apoptosis of human hepatocarcinoma cell in vitro.It suggests that TPT is a potent molecular drug for the treatment of patients with hepatocarcinoma.
