外周血单个核细胞Th类细胞因子表达与食管鳞状细胞癌的相关研究

摘　　要：目的 :探讨外周血单个核细胞 (PBMC)Th1 、Th2 类细胞因子的表达与食管鳞状细胞癌临床病理因素之间的相关性。方法 :采用双抗体酶联免疫吸附法 (ELISA)检测 39例食管鳞状细胞癌患者和 2 0例健康对照者外周血单个核细胞Th1 (IL 2、INF γ)、Th2 (IL 4、IL 6、IL 1 0 )类细胞因子的含量水平 ,并与食管鳞状细胞癌病理因素之间进行相关分析。结果 :食管鳞状细胞癌患者PBMC中Th1 类细胞因子IL 2、INF γ的表达明显低于健康对照组 (P <0 .0 5) ,Th2 类细胞因子IL 6的表达明显高于对照组 (P <0 .0 5) ,存在Th1 /Th2 漂移现象 ;且发现TNM分期越晚 ,Th1 类细胞因子IL 2的表达越弱 ,而IL 6表达越强 ,存在显著性差异 (P <0 .0 5)。Th1 /Th2 漂移现象与肿瘤分化程度无明显相关性。结论 :食管鳞状细胞癌患者PBMC存在某些Th2 类细胞因子表达优势 ,而Th1 类细胞因子表达减弱 ;Th1 /Th2Purpose: To study the relationship of the expression of Th 1/Th 2 type cytokines by peripheral blood in the patients with esophageal squamous cell cancer and its pathologic factors. Methods: Enzyme linked immunosorbent assay (ELISA) was used to detect the expression of Th 1(IL-2and INF-γ) and Th 2(IL-4, IL-6 and IL-10) type cytokine in 20 controls and 39 patients with esophageal squamous cell carcinoma.Results: Th 1/Th 2 shift exists in the patients with esophageal squamous cell cancer, levels of Th 1(IL-2 and INF-γ) type cytokines are significantly lower in the patients with esophageal squamous cell cancer than those in controls, but Th 2 type cytokines (IL-6) is on the contrary. In the late stage patients with esophageal squamous eel1 cancer,the level of Th 1 type cytokines (especially IL-2) is decreased, but IL-6 is more increased (P<0.05).Levels of Th 1(IL-2 and INF-γ) and Th 2 (IL-4, IL-6 and IL-10)type cytokines do not show relation with tumor differentiation. Conclusion: The level of Th 1 type cytokines were weaken in PBMC of patients with esophageal squamous cell cancer, but Th 2 type cytokines enhance. The Th 1/Th 2 shift are associated with TNM stage in patients with esophageal squamous cell cancer, but do not show relation with tumor differentiation.
