90-day subchronic toxicity study of aloe whole-leaf powder

摘  要:为了观察芦荟全叶粉长期食用对健康的影响 ,选用健康断乳SD大鼠 88只 ,随机分为 4组 ,每组 2 2只 ,雌雄各半 ,分别给予芦荟全叶粉 0、2、4、8g kgBW (按 0、2 5%、5%、10 %比例 )掺入基础饲料喂饲 90天。结果表明 ,芦荟全叶粉使大鼠排便量增加 ,粪便松软 ;雄性 8g kgBW剂量组大鼠体重增长明显缓慢 (P <0 0 1)、食物利用率降低 (P <0 0 1) ,4g kgBW剂量组食物利用率下降 (P <0 0 5) ,体重影响不明显 ,2g kgBW剂量组和雌性各剂量组对大鼠体重、食物利用率无影响。雄性 8g kgBW剂量组和雌性各剂量组肾 体比值增大 (P<0 0 1) ,4、8g kgBW剂量组睾丸 体比值增大 (P <0 0 5)。芦荟全叶粉对大鼠血液学指标 ,尿NAG酶 ,血清转氨酶、血脂、肌酐等生化指标无影响 ,8g kgBW剂量组血清BUN升高 (P <0 0 1)。病理检查 ,除 2g kgBW剂量组结肠粘膜无色素沉积外 ,各剂量组均可引起肾小管上皮、肠系膜淋巴结、结肠粘膜固有层色素沉积和肠系膜淋巴结增生 ,而肝、脾、睾丸 (或卵巢 )等脏器未见有意义的病理改变。本实验条件下 ,芦荟全叶粉有作用剂量为 2g kgBW (芦荟甙 11 8mg kgBW )。day subchronic toxicity study of aloe whole-leaf powder was conducted to ob serve the effect of aloe whole-leaf powder on health. 88 SD rats were divided r andomly into 4 groups, and each group consisted of 11 males and 11 females. The animals in group 2-4 received aloe whole-leaf powder mixed in regular rodent di et at doses of 2, 4 and 8 g/kg BW (by rate of 2 5, 5 and 10 percent in diet) for 90 days. The animals in group 1 received the regular rodent diet. The results s howed that aloe whole-leaf powder promoted defecation of rats. Food efficiency and body weight males exposed to 8g/kg BW were significantly less than those of the controls. Food efficiency males exposed to 4g/kg BW was significantly decrea sed, but body weight was not affected. No adverse effect were observed on food e fficiency and body weight by males exposed to 2g/kg BW and all exposed groups of females. Relative kidney weight was significantly increased in males exposed to 8g/kg BW and females exposed to 2g/kg BW or greater. Relative testis weight of rats exposed to 4 and 8g/kg BW were significantly increased compared to the cont rol groups. Aloe whole-leaf powder at the dose of 2, 4 and 8g/kg BW did not hav e adverse effect on hematology, serum AST、ALT、TC、TG、Cr as well as NAG activi ty of urine, serum BUN was significantly increased at the dose of 8g/kg BW. Pa thology findings: the incidences of pigmentation in renal tubular, mesenteric ly mph nodes and lamina propria of the colonic mucosa, and proliferation in mesente ric lymph nodes were significantly increased at all of the exposed groups, but n o pigmentation in colonic mucosa was observed at the dose of 2g/kg BW. No pathol ogical changes were observed in livers, spleens, testes (or ovaries). It was con cluded that observed adverse effect level of aloe whole-leaf powder was 2g/kg B W (LOAEL of aloin was 11 8g/kg BW).
