摘  要:研究水提、醇提及醇提-酸水解法不同提取工艺得到的决明子提取物对脂肪酶的抑制作用。采用脂肪酶抑制剂筛选模型对各提取物进行活性筛选,初步探究决明子提取物的抑制机理。决明子水提、醇提产物对脂肪酶无抑制作用,醇提-酸水解产物对脂肪酶有抑制作用,且在37℃、p H 7.4时抑制效果最佳,符合人体生理条件。Dixon图与Lineweaver-Burk图表明决明子醇提-酸水解蒽醌提取物对脂肪酶抑制作用为非竞争性抑制,抑制常数Ki=0.45 mg/m L。To study the extracts of aqueous extraction,alcohol extraction and alcohol extraction combined with hydrochloric acid hydrolysis by three different extraction processes and their inhibition effects on lipase,lipase inhibitor screening model to choose the active substance in Cassia obtusifolia L is adopted whcih can inhibit the activity of lipase,and preliminailx studied the inhibition mechanism on lipase of the Cassia obtusifolia L extracts. The result showed that extracts from aqueous extraction and alcohol extraction had no inhibitory effects on lipase,whife extracts from alcohol extraction combined with hydrochloric acid hydrolysis had inhibitory effects on lipase and the maximum inhibition rate at 37 °C,p H 7. 4 which was in line with human physiological conditions. Both the Dixon and the Lineweaver-Burk graphs showed that the extracts from alcohol extraction combined with hydrochloric acid hydrolysis have a noncompetitive inhibition on lipase,with the inhibition constant Ki = 0. 45 mg / m L.
