基质金属蛋白酶-2及其抑制物与大肠癌恶性生物学行为的关系

摘　　要：目的　探讨基质金属蛋白酶 -2 (MMP-2 )及其抑制物 (TIMP-2 )表达与大肠癌恶性生物学行为的关系。方法　采用 S-P免疫组化染色技术检测 3 0例大肠腺瘤、60例大肠癌组织 MMP-2和 TIMP-2的表达情况。结果　MMP-2的表达率在大肠腺瘤中为 2 6.7% (8/ 3 0 ) ,显著低于大肠癌中的 86.7% (52 / 60 ) (P<0 .0 5) ;其表达与大肠癌的 Dukes分期、分化程度、淋巴结转移和生存期均密切相关 (P值均 <0 .0 5) ;本组大肠癌 Dukes B期比例较高 ,其总体 TIMP-2表达在腺瘤和本组大肠癌之间差异无显著性 (P>0 .0 5) ,TIMP-2表达与大肠癌的 Dukes分期、淋巴结转移、远隔脏器转移及生存期均有关 (P值均 <0 .0 5)。结论　 MMP-2和 TIMP-2的检测可作为临床判断大肠癌的恶性程度。ObjectiveTo investingate the relationship of the expression of matrix metaloproteinases- 2(MMP-2) and its tissue inhibitor (TIMP-2) with malignant biological behavior of colorectal cancer(CRC). MethodsThe MMP-2 and TIMP-2 expr ession was studied with immunohistochemistry in 30 specimens of colorectal adeno ma and 60 specimens of CRC. ResultsThe expression rates of MMP -2 were 26 7 in colorectal adenoma and 86 7% in CRC (P<0 05). The signif icant relationship was observed between high MMP-2 expression and Dukes stage, cellu lar differentiation, metastasis of lymph nodes and poor prognosis. No significan t difference was observed in expression of TIMP-2 between colorectal adenoma an d CRC (P0.05). The significant relationship was observed between low TIMP- 2 expression and Dukes stage, metastasis of lymph nodes and distant organs, and poor prognosis. ConclusionThe detection of expression of MMP -2 and TIMP-2 may be helpful to indicate the malignant behavier, metastasis an d prognosis in colorectal carcinoma.
