摘　　要：目的 :探讨人重组骨形成蛋白 2 (recombinanthumanbonemorphogeneticprotein 2 ,rhBMP 2 )对舌癌细胞Tca8113增殖能力的影响 ,以深入了解口腔舌癌的恶变机制。方法 :用含 10 0ml/L胎牛血清的RMPI 164 0培养基 ,加入 5 0 μg/L的rhBMP 2培养舌癌细胞Tca8113 ,通过细胞生长曲线、MTT检测、流式细胞仪分析观察细胞的生长状态 ,免疫组化检测细胞周期蛋白依赖性激酶 4(cyclin dependent kinases 4,CDK 4)的表达来观察细胞的周期变化。 结果 :5 0 μg/L的rhBMP 2能显著增进Tca8113细胞的增殖 ,免疫组化结果显示周期蛋白CDK 4的表达增强。结论 :5 0 μg/L的rhBMP 2能显著促进Tca8113细胞的增殖 ,并上调细胞周期蛋白的表达 ,说明BMP 2可能在舌癌的发生发展过程中起着重要作用。Objective: To examine the effects of rhBMP-2 on the proliferation of tongue cancer Tca8113 cells and on the expression of CDK-4 in the cells. Methods :Tca8113 cells were cultured in RMPI 1640 containing 50 μg/L rhBMP-2. The proliferation of the cells was investigated by cell counting, MTT assay and flow cytometry. The expression of CDK-4 of the cells was investigated by immunohistochemistry method. Results: Tca8113 cells cultured in RMPI 1640 containing 50 μg/L rhBMP-2 increased the proliferation of the cells (P<0.05) , increased proportion of S and G 2 phase cells and induced stronger expression of CDK-4 (P<0.05) of the cells. Conclusion: rhBMP-2 may stimulate the proliferation of oral squamous carcinoma cells.
