松针褐斑病菌毒素的层析分离及紫外吸收特性研究  被引量:3

摘  要:以生测为导向,采用柱层析和薄层层析相结合,从松针褐斑病菌发酵液中分离出3个致毒活性组分m,p和q。经薄层层析检验可知m,q在A,C两个展开系统中Rf值一致,分别为0.57和0.75,且m,q展开后在254nm和365nm紫外光下都呈蓝色荧光,基本认为m和q是同一组分;p在365nm紫外光下为黄白色荧光,在A、C两个展开系统中Rf值分别为0.46和0.66。层析分离的结果表明,松针褐斑病菌可能含两个或两个以上的毒素物质。对m和p两组分进行紫外扫描的结果显示,m,p分别在波长210nm和208nm处有最大吸收峰,在365nm和254nm紫外光下无吸收或吸收很弱。高效液相分析显示:m,p都不是纯物质,在210nm检测波长下,m有3个吸收峰,保留时间分别为2.41,3.80和16.68min,p有4个吸收峰,保留时间分别为2.46,3.75,12.00和17.25min。研究结果表明组分m和p可能含有相同或相似的化学物质,要获得纯物质还需进一步分离。Lecanosticta acicola that is pathogenic fungus of brown spot needle blight could produce pathogenic toxin. Leaded by bioassay,three active fractions(m,p and q) were obtained from metabolites of the pathogen in MS culture liquid by the separation way of combination of silica column chromatography(SCC) and tiny layer chromatography(TLC).Both m and q showed somely blue fluoresent light under 254 nm and 365 nm UV light on TLC plate and had the same Rf values developed by two solvent systems(A and C),which were 0.57 and (0.75) respectively.Active fraction p fraction was yellow-whited fluorescence under (365 nm) and the Rf values were 0.46 and 0.66 developed by A and C,which were different from the Rf values of m and q.These results indicated that two or more toxins would exist in the metabolites of Lecanosticta acicola.
