摘 要:目的:研究咖啡因(CAF)对DDP的抗癌增效作用及苯巴比妥(PBT)是否对咖啡因的抗癌增效作用产生抑制。方法:以人宫颈癌细胞ME-180为靶细胞,采用MTT法研究甲基黄嘌呤类药物-咖啡因及咖啡因与苯巴比妥联合应用对DDP的体外细胞毒作用的影响。结果:120μg/mlCAF及10μg/mlPBT对ME-180的生长无明显细胞毒作用。120μg/mlCAF可明显增强DDP对ME-180的细胞毒作用(P<0.05),10μg/mlPBT对DDP无增效作用(P>0.05),120μg/mlCAF与10μg/mlPBT的混合物也能增强DDP的细胞毒作用(P<0.01),其增效作用与单纯CAF的增效作用比较无显著性差异(P>0.05)。结论:咖啡因可明显增强DDP对ME-180细胞的体外抗癌效应,苯巴比妥对咖啡因的体外抗癌增效作用无拮抗作用。提示苯巴比妥作为咖啡因的减毒成分与咖啡因联合共同作为化疗增效剂用于宫颈癌治疗的可能性。Objective:To investigate the enhancing effect of caffeine (CAF)on the anti-cancer activity of cisplatin of human ME-180cells and if this enhancing effect of caffeine could be inhibited by phenobarbital(PBT).Methods:MTT assay was used to determine the effect on the cytotoxicity of DDP in ME-180cell by caffeine,a methyxanthine derivative,and the combination of caffeine plus phenobarbital.Results:No significant cytotoxicity was observed in ME-180cell treated with120μg /ml CAF and10μg /ml PBT alone or combinated.120μg /ml CAF showed sig-inificant enhancing effect on the cytotoxicity of DDP in ME-180(P<0.01)and10μg /ml PBT showed no enhancing effect(P>0.05),while120μg /ml CAF+10μg /ml PBT also showed enhancing effect of the cytotoxicity of DDP(P<0.01)which had no difference with that of10μg /ml CAF(P>0.05).Conclusion:Caffeine could enhancing anticancer activity of DDP in ME-180.This enhanc-ing effct of caffeine was not antagonized by Phenobarbital.This suggests the possibility of peno-barbital as a composititon to reduce the cytotoxicity of caffeine in vivo and together with caffeine as an enhancing agent of chemotherapeutical drugs to against cervical cancer.
