摘  要:目的:探讨IL-27对葡聚糖硫酸钠(Dextran sodium sulfate, DSS)诱导的小鼠溃疡性结肠炎及IL-12家族成员(IL-12, IL-23, IL-27, IL-35)表达水平的影响。方法:用DSS自由饮用法复制Balb/c小鼠溃疡性结肠炎模型。实验设对照(NC)组、模型(UC)组、IL-27高剂量(H)组、低剂量(L)组。造模后,UC组,H组和L组分别予生理盐水、IL-271 μg/20g/天和0.2 μg/20g/天皮下注射治疗4 d,观察一般情况并收集血清和结肠组织标本。用ELISA法检测小鼠血清IL-12、IL-23、IL-27、IL-35的水平,用RT-PCR法检测小鼠结肠组织中IL-27P28、IL-27EBI3、IL-12P19、IL-23P35的相对含量。结果:DAI评分、MPO水平明显增高(P < 0.05);与UC组相比,H组、L组明显降低(P < 0.05);小鼠体重、结肠长度、重量及脾脏重量明显降低(P < 0.05),与UC组比较,H组和L组增高(P < 0.05)。ELISA结果:IL-12、IL-23水平增高(P < 0.05);与UC组比较H组、L组明显降低(P < 0.05);与NC组比较,IL-27的表达在UC组稍有增高,H组,L组明显增高(P < 0.05);与UC组比较,H组,L低明显增高(P < 0.05);与NC组比较,IL-35的表达在UC组明显降低,H组,L组有所降低升高(P < 0.05)。RT-PCR检测:IL-27P28和IL-27EBI3的mRNA相对含量:与UC组比较,H组和L组明显增高(P < 0.05);而IL-12P35与IL-23P19的mRNA相对含量显著低于UC组(P < 0.05)。结论:IL-27对UC有防护作用,其机制可能与抑制IL-12、IL-23,促进IL-27、IL-35的表达有关。Objective: To study IL-27 treatment on dextran sodium sulfate (DSS)-induced ulcerative colitis in mice and the expression of IL-12 family members (IL-12, IL-23, IL-27 and IL-35). Methods: Balb/c mice ulcerative colitis model was induced using DSS preparation. The mice were divided into con-trol group (NC), model group (UC), IL-27 high dose group (H), IL-27 low dose group (L). UC group to the physiological saline, H and L groups respectively to the IL-271 μg/20g/day, IL-270.2 μg/20g/day subcutaneous injection treatment for 4 days, observe the general situation and collect serum and colon tissue specimens. with ELISA method to detect the level of IL-12, IL-23, IL-27, IL-35int serum;by RT-PCR method detecting the mRNA relative content of IL-27EBI3, IL-12P19, IL-23P35 in the colon tissue in mice. Results: DAI, MPO level increased significantly (P < 0.05);H and L group compared with UC group significantly decreased (P < 0.05);Body weight in mice, colon length, weight and spleen weight significantly decreased (P < 0.05), compared with UC group, H and L group increased (P < 0.05). ELISA results: IL-12, IL-23 levels increased in UC group (P < 0.05);H, L group IL-12, IL-23 levels significantly decreased (P < 0.05);Compared with NC group, the expression of IL-27 in UC group slightly increased;H, L group significantly increased (P < 0.05);Compared with NC group, the expression of IL-35 in the UC group significantly decreased, H, L group significantly increased (P < 0.05). Rt-pcr detection: IL-27P28 and IL-27EBI3 mRNA relative content: compared with UC group, H and L group significantly increased (P < 0.05);IL-12P35 and IL-23P19 mRNA rela-tive content was significantly lower than UC group (P < 0.05). Conclusion: IL-27 of UC has protective effect, its mechanism may be related to inhibition of IL-12, IL-23, promoting the expression of IL-27 and IL-35.
