摘  要:以大豆油脚为原料 ,采用溶剂萃取法和柱色谱法相结合的工艺制备高纯卵磷脂和脑磷脂 ,研究了不同洗脱剂及不同洗脱模式对卵磷脂和脑磷脂柱色谱分离效果的影响。实验结果表明 :大豆油脚先经脱水、脱油 ,再经 95 %乙醇浸提后粗分为粗卵磷脂和粗脑磷脂 ,以硅胶为吸附介质 ,分别进行柱色谱分离。粗卵磷脂为原料用无水甲醇作为洗脱剂等度淋洗 ,可获得大量相对含量为91.9%的卵磷脂和少量含量为 74 .2 %的脑磷脂产品 ;粗脑磷脂为原料用氯仿 -甲醇 ( 2∶1,V/V)、无水甲醇的梯度淋洗模式 ,可获得少量含量 96 .7%的高纯卵磷脂和大量含量 82 .1%的脑磷脂产品。The high purity soybean PC and PE were prepared with combining methods of solvents extraction and column chromatography from soybean oil degummed residues. The effects of different eluents and eluenting modes on separation of PC and PE were also studied. The soybean oil sediment was removed water and oil, and then separated into two parts (crude PC and PE) by using 95% ethanol extraction as the raw materials for column chromatography separation. A great deal of high purity PC(91.9%) and a little of PE(74.2%) can be obtained by using column chromatography of silica as sorbent with isocratic eluenting of methanol from crude PC. A little of high purity PC(96.7%) and a great deal of PE(82.1%) can be also obtained by gradient eluenting with chloroform-methanol (2∶1,V/V) and methanol from crude PE.
