产脂肪酶菌株的筛选及紫外-光复活诱变

摘　　要：目的从土壤中分离选育出脂肪酶高产菌株。方法通过罗丹明B指示平板法分离出产脂肪酶量高的菌株,通过紫外-光复活诱变选育提高产量。结果从土壤中筛得脂肪酶产量为6.75 U/mL的菌株白地霉L-8,通过紫外-光复活诱变提高产量到12.08 U/mL。结论获得了一株高产脂肪酶的白地霉菌株,结果表明从土壤中分离出产脂肪酶的菌株,再通过紫外-光复活诱变提高其产量的高产菌株的筛选方法是可行的。Objective To isolate and select a high-yield lipase producing strain from soil.Methods A high-yield strain was isolated by flat separation using Rhodamine B as indicator.Its production was improved by ultraviolet photoreactivation mutagenesis.Results We obtained Geotrichum candidum L-8 from soil.Its lipase yield was improved from 6.75 U/mL to 12.08 U/mL through ultraviolet photoreactivation mutagenesis.Conclusion A high-yield lipase producing strain was obtained.The methods to isolate it from soil and to improve its production of lipase were feasible.
