摘 要:利用PCR-单链构型多态性(PCR-SSCP)技术检测75头乌金猪和41头长白猪脂肪细胞定向和分化因子1(ADD1)基因第2外显子区的遗传变异,并用最小二乘法模型分析ADD1基因对乌金猪和长白猪肌内脂肪含量的遗传效应。结果表明:在所检测的样本中均在乌金猪和长白猪ADD1基因的第2外显子上发现多态性,有2个等位基因(A和B),2种基因型(AA和AB),缺少BB基因型。AA和AB基因型在乌金猪中的频率分别为0.240和0.760,在长白猪中的频率分别为0.634及0.366。两种猪的优势等位基因均为A,其基因频率在乌金猪和长白猪分别为0.620和0.820。通过对两种猪不同基因型与肌内脂肪含量的相关分析,发现ADD1基因多态性对乌金猪和长白猪肌内脂肪含量的影响显著,且AB>AA(P<0.05),可通过提高AB基因型的频率来增加肌内脂肪含量,达到改善猪肉品质的目的。Polymorphism of the extron 2 on adipocyte determination and differentiation factor 1(ADD1) gene of 75 Wujin pigs and 41 Landrace were tested by means of PCR-single strand conformation polymorphism(PCR-SSCP),and the relations of ADD1 gene to intramuscular fat contents(IMF) were analyzed by general linear model(GLM).The results showed that the polymorphism existed in two kinds of pigs on ADD1 gene and there were 2 alleles(A,B) and 2 genotypes(AA,AB).The genotype frequencies of AA,AB of ADD1 gene in Wujin Pig were 0.240 and 0.760 as well as 0.634 and 0.366 in Landrace,respectively.Allele A was dominant gene and its frequency in two pigs was 0.620 and 0.820,respectively.The IMF of two pigs were affected significantly by ADD1 gene and showed a trendy of AB > AA(P<0.05).It is suggested that the ADD1 gene can be acted as a candidate marker for selecting IMF content in pigs.
