黄桃水溶性多糖的化学结构分析

摘  要:采用水提醇沉法从黄桃果肉中提取出水溶性多糖HTP,HTP先后经DEAE-Sepharose离子交换色谱和Sephacry1G-300凝胶过滤色谱纯化得到HTP1和HTP2,经高效分子排阻色谱法(简称HPSEC)鉴定其纯度及计算分子量,HTP1和HTP2均为单一多糖,分子量分别为52107和59855。HTP1和HTP2经完全酸水解后,分别经TLC和HPLC分析,发现HTP1单糖组成为D-Glc、D-Gal、D-Arb和L-Rha,其摩尔比为1.0∶4.2∶14.5∶1.5;HTP2单糖组成为D-GalA、D-Man和D-Gal,其摩尔比为34.3∶5∶1。经高碘酸氧化、Smith降解、紫外光谱、红外光谱和核磁共振等方法分析其连接方式和构型,结果表明,HTP1多糖主链为α-(1→3)键连接的阿拉伯半乳糖结构,侧链含有少量的D-Glc和L-Rha;HTP2多糖主链为α-(1→3)键连接的D-GalA,侧链含有部分D-Man和D-Gal。The water-soluble polysaccharides of peach were extracted with hot water, and then isolated by ethanol. HTP1 were further purified to HTP1 and HTP2 with DEAE-Sepharose ion exchange column chromatography and Sephadex-300 gel filtration column chromatography, respectively. The high performance size-exclusion chromatography proved that HTP1 and HTP2 were pure material. The average molecular weight of HTP1 was 52107 and the average molecular weight of HTP2 was 59855. Their structures were investigated by chemical methods (composition analysis, periodate oxidation, and Smith degradation) and spectroscopic methods (UV, IR, NMR). HPLC and TLC identified their monosaccharide after TFA hydrolyze. HTP1 is composed of D-glucose, D-galactose, D-Arabinose and L-Rhamnose in the ratio of 1.0∶4.2∶14.5∶1.5,HTP2 is composed of D-galactose, D-Mannose and D-Galacturonic acid in the ratio 34.3∶5∶1. The result of periodate oxidation and Smith degradation showed that the major linkage of HTP1 is (1→3) D-galactose and D-Arabinose. The major linkage of HTP2 is (1→3) D-Galacturonic acid. Infrared analysis showed that HTP1 and HTP2 were all α-glycoside linkage.
