摘 要:用壳聚糖絮凝除去十五碳长链二元酸发酵液中的酵母菌.当壳聚糖浓度为600μg·g-1,温度为30℃,pH为8.5~9.0时,离心除去菌体后,上清液透光度可提高5倍以上,同时可除去78%的蛋白质.文中还讨论了絮凝体去除方法的选择,结果表明,用低速离心或转鼓过滤时,二元酸损失量较小,二元酸收率达88%.用壳聚糖絮凝除去菌体及蛋白是一种简单实用的二元酸分离纯化方法.Flocculation of yeast cell from broth of 1, 13-dicarboxylic acid with chitosan was studied. When the concentration of chitosan was 600 μg. g-1, the temperature was 30℃ and the pH was 8. 5~9. 0; the turbidity of the supernatant after centrifugation decreased to one fifth of the original. The 78% of the proteins was also removed by flocculation. The result indicated that low speed centrifugation or rotary drum filter should be used in the removal of cell to reduce the loss of dicarboxylic acid. The flocculation of cells with chitosan is a simple and inexpensive method for the isolation and purification of dicarboxylic acid.
