摘  要:龟鳖类同其它类群脊椎动物的系统进化关系一直存在争论。为进一步从分子水平上探讨这一问题,本文参照近源物种的线粒体基因组,设计了16对特异引物,采用PCR产物直接测序法测得了乌龟线粒体基因组全序列。结果表明:乌龟线粒体基因组序列全长16576bp,包括2个rRNA基因、22个tRNA基因、13个蛋白质编码基因和1个非编码控制区。乌龟线粒体基因组结构和基因排列顺序与其它龟鳖类相同,在“WANCY区”包含一个“stemloop”结构,ND3基因174位点存在一个额外插入的腺苷酸(A)。本文通过比较分析结构基因在主要脊椎动物类群中的排列顺序。The phylogenic relationship of turtles to other vertebrate groups remains to be open. In order to provide molecular evidence to this issue, we analyzed the complete mitochondrial (mt) genome of the turtle Ckinernys reevesi. DNA sequencing was based on PCR fragments from 16 primer pairs that were designed on the basis of mtDNA sequences of three related turtle species. The genome is 16576 base pairs in length, contains 37 genes (two ribosomal RNA, 22 transfer RNA and 13 protein-coding genes) and a putative control region, a situation that is similar to that found in other vertebrates. The gene order is identical to that of the four other turtle species so far reported. Those genes are arranged compactly with no introns and few intergenic nucleotides. Twenty-two tRNA genes include 2 tRNA^Ser, 2 tRNA^Leu and 18 different species of other tRNA genes each. The tRNA^Gln, tRNA^Ala, tRNA^Asn, tRNA^Cys, tRNA^Tye, tRNA^Ser (the second one) , tRNA^Glu and tRNA^Pro genes are encoded on the light strand, and the remainder is on the heavy strand. The length of the tRNA genes ranges from 66 bp (tRNA^Ser and L-strand) to 77 bp (tRNA^Ttp). All protein-coding genes begin with ATG as initiation codons except CO1 that uses GTG. Most protein-coding genes, including CO2, ATP8, CO3, ND4L, ND4, ND5 and Cyt b, have a TAA termination codon, while the others have incomplete termination codons. Among the 13 protein-coding genes, only ND6 is located on the light strand, the others are on the heavy strand. The order of the 13 protein-coding genes is the same as in mammals, amphibians and crocodiles. The total length of the 13 protein-coding genes makes up 11378 bp, which is 68.64% of the mt genome. The 12S rRNA gene of Chinemys reevesi begins at 73 bp and ends at 1041 bp. The 16S rRNA gene begins at 1112 bp and ends at 2715 bp. The control region of the Chinemys reevesi mt genome is 1072 bp long, which is 68.66% AT-rich region, and its flanking tRNA genes are 5'-tRNA^Pro and 3'-tRNA^Phe. The sequence was submitted to G
