摘  要:研究了几种有机溶剂对南美白对虾(Penaeus vannamei)N-乙酰--βD-氨基葡萄糖苷酶(NAGase)活力的影响.结果表明:甲醇、乙醇、正丙醇、正丁醇、乙二醇和丙三醇对NAGase的作用均表现为低浓度时对酶有激活作用,随着浓度的升高,激活作用减弱并速转为抑制作用;二甲亚砜和二氧六环对酶的抑制作用呈现明显的浓度效应,随浓度增大酶活力迅速下降.进一步研究二甲亚砜的抑制作用动力学,结果表明:二甲亚砜对酶的抑制作用是一种可逆过程,抑制机理表现为混合型抑制类型,抑制常数KI=0.27 mol/L,KIS=2.05 mol/L.Effects of different organic solvents on β-N-Acetyl-D- glucosaminidase (EC3. 2.1. 52) from Penaeus vannamei was investigated. The results indicated that the enzyme was activated in methanol, ethanol, propanol, glycol and glycerol solutions at low concentration, but inhibited at high concentration. The effects of dimethyl sulfoxide and dioxane on the enzyme were concentration-dependent. As the concentrations of dimethyl sulfoxide and dioxane increased,the residual enzyme activity rapidly decreased. The concentrations of dioxane and dimethyl sulfoxide leading to 50 % of the enzyme activity lost, IC50 ,were determined to be 1.05 and 1.12 mol/L. The inhibitory kinetics of dimethyl sulfoxide on the enzyme was further studied. The result showed that the inhibition reaction of the enzyme in dimethyl sulfoxide solvent was a reversible process and the inhibition type was determined by Lineweaver-Burk plots to be mixed-type. The inhibition constants K1 and K1s were determined to be 0.27 mol/L and 2.05 mol/L,respectively.
