摘 要:里氏木霉(Trichodermareesei)A_3经亚硝基胍和紫外线复合处理,获得一株纤维素酶高产菌株91-3。该菌株在最适固态发酵条件下,纤维素酶滤纸酶活力为170u/g曲,产酶水平是出发菌株的1.6倍。酶作用的最适条件为pH4.8,50℃;pH稳定范围为3~7;90℃处理7min,酶活保存率为91.64%;室温放置半年,酶活保存率在90%以上,室温放置一年,酶活保存率在80%以上。mutant strain 91-3 with high potency for cellulase production was obtained from Trichoderma reesei A3 which was treated with UV and NTG at room temperature.Cellulase Qu was obtained by growing the mutant strain on semi-solid state medium consisted of 3.5g of maize straw powder,1.5g of wheat bran,15ml of nutrition salt solution in 250ml of flasks at 30℃ for 96h. Enzyme activity of the Qu was 170u/g on filter paper.Cellulase activity was about 1.6 times that of the parent strain.The optimal conditions for enzyme action on filter paper were PH4.8 and 50℃.The enzyme was stable at PH 3.0~7.0.After incubating the enzyme at 90℃ for 7mins,90% of the enzyme activity remained.Put the enzyme at room temperature for one year, 80% of the enzyme activity remained.
