摘  要:目的:探讨海藻氨酸(Kainicacid,KA)致痫后海马神经元谷氨酸转运体(glutamatetransporters,GluTs)功能的变化,并观察牛磺酸(Taurine,Tau)对KA致痫及致痫后海马GluTs功能的影响。方法:40只Wistar大鼠随机分为对照组(Ⅰ组)、KA组(Ⅱ组)、Tau低剂量组(Ⅲ组)、Tau高剂量组(Ⅳ组),每组各10只。Ⅲ组和Ⅳ组提前3d分别腹腔注射低剂量(1g/kg)和高剂量(2g/kg)Tau,然后与Ⅱ组一起腹腔注射KA10mg/kg,诱导癫痫发作,Ⅰ组腹腔注射生理盐水。观察各组的痫性发作情况,并于24h后剥离右侧海马,制备海马突触颗粒,测定其摄取3HL谷氨酸的功能。结果:与对照组相比,KA组点燃后海马突触颗粒GluTs功能降低(P<0.01);与KA组相比,牛磺酸组的致痫潜伏期延长,点燃率、痫性发作分级及死亡率均降低,海马神经元GluTs功能增强,其作用呈剂量依赖性。结论:KA可造成海马神经元GluTs功能降低,Tau抑制KA引起的癫痫发作,其原因可能与部分改善海马神经元GluTs功能有关。Objective: To study the change of glutamate transporters (GluTs) function in hippocampus neuron of kainic acid (KA) kindled rats, and observe the influence of taurine (Tau) on the kindling and the function of GluTs. Methods: Fourty Wistar rats were divided randomly into 4 groups: Ⅰ group(saline control group), Ⅱ group ( KA group), regroup ( low dose taurine group), Ⅳ group ( high dose taurine group), Ⅲ function by means of assay of ^3H-L- glutamate uptaken to control group, the GluTs function of KA group was in synaptosomes of hippocampus. Results: Comparing significantly decreased (P〈0.01). Comparing to KA group, the onset latency of taurine group increased; the occurrence of tonic seizures, the grade of seizure behavior and the mortality rate reduced; and the function of GluTs ameliorated, which depended on the dosage of taurine. Conclusion: KA can cause GluTs function decreasing in hippocampus neuron; taurine can prevent the seizures induced by KA; this partly contributes to the amelioration of GluTs function in hippocampus neuron.
