金针菇的深层液体培养及应用

摘　　要：筛选菌株的发酵曲线表明,培养48～96h为其增殖生长期;菌龄试验表明,80h摇瓶菌种活力优于100 h和130 h者;发酵罐中试培养中,65h生物量即达33.7 g.mL-1(鲜重);醪液作为液体菌种,产量及菇形正常,发菌期缩短1/3,且现蕾期较早,从醪液中可提取营养浓缩液及粗多糖粉。浓缩液含氨基酸555.38mg.100mL-1,多糖含量≥6.0 mg.mL-1,可溶性固形物≥13%,急性毒性试验证明无毒,重金属含量符合国家标准。动物实验证明其具有显著的免疫活性。The fermentation curve of the selected strain of Flammulina velutipes indicates that the multiplication period is 48～96 hours.The activity of the shake flask spawn of 80hours is better than that of 100 hours and 130 hours.The biomass is 33,7g/100ml （fresh weight） at 65 hours in the fermenter.The spawn running time reduces by one-third and primordiun formation is earlier than that of the nomal cultivation when mash is used as liquid spawn. Concentrated nutrient liquid and coarse polysaccharide meal can be extracted from the mash.And the content of amino acid, polysaccharide and soluble solid is 555.38mg/100ml, 6.0mg/ml and≥13% resp.in the concentrated liquid. The concentrated liquid is proved to be safe by the acute toxie test and the content of heavy metal is in accordance with the national standard. Animal tests showed that the effect of F, velutipes nutrient solution on the organism immunity is extremely obvious.
