膳食多不饱和脂肪酸构成对小鼠淋巴细胞功能及脂肪酸构成的影响

摘  要:目的观察膳食n-6/n-3PUFA比值对淋巴细胞脂肪酸构成和细胞功能的影响。方法48只BALB/c小鼠随机分为4组,即n-6/n-3比值分别为1,7.5,15,30;基础饲料采用AIN-93G配方,S:M:P为1:1.5:3.7。饲养12周。测定小鼠T淋巴细胞功能、IL-2水平和淋巴细胞脂肪酸构成。结果n-6/n-3比值接近1时,小鼠T淋巴细胞增殖活性及培养上清IL-2水平显著降低;淋巴细胞C18:3,C20:6,n-3PUFA中含量显著减少;淋巴细胞C22:6含量与淋巴细胞增殖活性显著负相关,C18:3,C20:5,C22:6含量与CD4+T淋巴细胞比例显著负相关;淋巴细胞脂肪酸构成的改变基本与膳食脂肪酸的改变基本一致。结论膳食S:M:P为1:1.5:3.7时,n-6/n-3比值为1时,小鼠T淋巴细胞增殖活性受到抑制,其作用可能与IL-2的降低有关;小鼠脾淋巴细胞的脂肪酸构成受膳食脂肪酸构成的影响。Objective To study the effect of dietary n-6/n-3 PUFA ratio on function of spleen lymphocyte and fatty acid composition in mice.Methods 48 male BALh/c mice were divided into 4 groups, according to the n-6/n-3 PUFA ratio of 1, 7.5, 15 and 30, respectively. The dietary S:M:P was 1 : 1.5:3.7 shmilar to AIN-93G formula. After 12 weeks of breeding, all mice were sacrificed. The function and fatty acid composition of lymphocyte, IL-2 concentration were raeasured. Resuits When dietary n-6/n-3 ratio approximated 1 , the lymphocyte proliferatian and IL-2 level significantly decreased; The concentration of C18:3,C20:6 and n-3 PUFA in lymphocyte significantly decreased; The concentration of C22:6 in lymphocytes was negatively correlated with the proliferation of spleen lymphocytes. The concentration of C18:3, C20:5 and C22:6 in lymphocyte was negatively correhted with the proportiom of CD4^+ T cell. Conclusion In diet with 1 : 1.5:3.7 of S: M: P, T lymphocyte function of mice is suppressed when dietary n-6/n-3 ratio approximates 1, which might be related to decrease in IL-2. The fatty acid composition of lymphocyte is affected by diets in mice.
