摘 要:对胡芦巴中性杂多糖酶解的工艺条件及其产物对青春双歧杆菌增殖作用进行研究。以还原糖得率为参考指标,采用L9(34)正交设计对反应温度、反应时间、酶量的参数进行优选,观察了水解效率不同的两种方案的酶解产物及其中不同分子量的组分对青春双歧杆菌的增殖效应。结果表明,方案5即反应温度50℃、反应时间9 h和酶量6%,最有利于胡芦巴中性杂多糖的酶解,其产物更有利于双歧杆菌增殖,该方案的胡芦巴中性杂多糖酶解产物经Sephadex G-150柱层析,得到两个主要组分,其均分子量分别为4.5×104和7.5×103,并证实分子量较小的多糖酶解片断更有利于青春双歧杆菌的增殖。This paper studied the enzyme hydrolysis conditions of fenugreek neutral heteropolysaccharide and the effects of its products on reproduction of Bifidobacterium adolescentis. The orthogonal test was used to optimize the reaction temperature, reaction time and enzyme content, using the yield of oligosaccharide as an index, the products of two schemes with different hydrolysis efficiency and different molecular weight components, showed significant effects on the growth of B. adolescentis. The results showed that the No. 5 scheme (9 h reaction time, 50 ℃ reaction temperature and 6% enzyme content) was the optimal condition for enzyme hydrolysis of fenugreek neutral heteropolysaccharide and its products had better effects on the growth of B. adolescentis. Molecular weights of two components, which were from the products separated by Sephadex G-150, were 4. 5 × 10^-4 ,7.5 × 10^3 respectively, and the small one had better effects on B. adolescentis growth.
