碱性淀粉酶产生菌的筛选和产酶条件的研究

摘　　要：对采自全国各地的５０份上样进行筛选，获得一株产酶活性较高的菌株Ｂ－９５４５，通过摇瓶培养确定了产酶的最适培养基Ⅱ（ｇ／１００ｍｌ）：复合蛋白胨１．０；玉米淀粉１．５；玉米浆１．５；ＮａＣ１０．５；Ｋ２ＨｐＯ４０．１；ＭｇＳＯ４０．０２；ＣａＣＯ３０．５；Ｔｗｅｅｎ８００．１；ｐＨｇ—１０，在该条件下，３０℃荡培养４８ｈ，酶活力可达５９６ｕ／ｍｌ。对菌株Ｂ—９５４５的产酶特性研究表明：该酶对高ｐＨ环境有较强的适应性，因此，可用于碱性洗涤用酶。A wild strain B-9545 with high alkaline amylase activity is isolated after screening 50 samples of soil from all over the country. In shake flask calture, the optimum conditions for enzyme production are in the medium Ⅱ Containing(g/100ml ): Polypeptone 1.0; Corn starch 1. 5; Corn steep Liquor 1. 5; NaCl 0. 5; K2HPO4 0.1; MgSO4 0. 02; CaCO3 0. 5;Tween 80 0. 1; pH 10. Under such conditions, incubated at 30℃. for 48h,its enzyme activity can reach 596u/ml.Some tests about the properties of the alkaline amylase Produced by strain B-9545 show that it has strong tderance to high pH and may be used as detergent indus try Zneyme.
