摘  要:花生四烯酸(AA)是一种重要的人体必需多不饱和脂肪酸(PUFA),具有广泛的生物活性。将真菌油脂皂化制得的混合脂肪酸经尿素包合法纯化和甲酯化后,再利用负载在氧化铝上的氯化亚铜(CuCl-Al2O3)(质量比为1:1)为固定相,以含1%丙酮的正己烷溶液(体积比)为流动相的柱色谱法进一步纯化其中的花生四烯酸,流速1~1.5mL/min,负载量0.52g/100gCuCl-Al2O3,结果使花生四烯酸含量由74.9%提高到93.5%,收率为80.5%。采用气相色谱(GC)进行跟踪检测分析。Arachidonic acid ( AA ), an importantly essential polyunsaturated fatty acid ( PUFA ) to human nutrient, has broad biological activities. The PUFA ,which obtained from saponified fungal oil, was isolated by urea inclusion firstly ,and then methylated. The PUFA methyl esters was further purified via Al2O3-supported CuCl (l: 1, W/ W) chromatography under the following conditions:the eluent 1% acetone in hexane ( V/V) , the flowing rate 1 - 1.5 mL/min,and the loading amount 0. 52 g/100 g CuCl-Al2O3. The product of high purity which contains 93.5% AA with 80. 5% yield was obtained from the PUFA methyl esters containing 74. 9% AA. The process was monitored and analyzed by GC.
