一株琼胶酶高产菌株的筛选鉴定及产酶条件的优化

摘　　要：本研究从海水中分离筛选到一株琼胶酶高产菌,编号是2.2-g,革兰氏染色为阳性,呈细杆状。通过DNS法测定,其分泌琼胶酶的酶活为0.066U。通过16SrDNA的PCR扩增、序列测定,测序结果(AY949021)与Genebank数据库进行比较,发现它与Brevibacteriumaureum(AY299093)和Brevibacteriumlinens(AY243345)同源性高达99%,因此可以判断其为短杆菌(Brevibacteriumsp)。A straight Gram-positive bacteria containing highyield agarase was selected from sea water and named 2.2-g. The enzyme activity of agarase isolated from the bacteria was 0.066 U detected by DNS method. By means of 16S RNA analysis and compared the analysis results with Genebank database, it had 99% homology to Brevibacterium aureum （AY299093）, and Brevibacterium linens （AY243345）. It belonged to Brevibacterium sp.
