摘 要:Odontogenic differentiation and precise mechanism of dentinogenisis have not been well characterized partly because of the limited life span of native odontoblast. The ubiquitous transcription factor c-Myc, encoded by Myc proto-oncogene, is believed to play an important role in cell immortalization and cancerization by upregulating telomerase activity in some kinds of human somatic cells. The purpose of this study is to understand whether the telomerase activity could be regulated by c-Myc in human odontoblast. Primary human odontoblast-like cells were cultured and transfected with c-Myc expressing vector. After selection with G418 for one month, c-Myc was integrated and expressed stably at protein level, and telomerase activity and expression of human telomerase reverse transcriptase (hTERT) were upregulated in these transfected cells." Thus, telomerase could be activated by exogenous c-Myc through upregulation of hTERT expression in human odontoblast-like cells. This laid a foundation for establishing immortalized human odontoblast cell line.
