摘  要:目的选取当前市场销售的品质不一的10种海参,分析比较其多糖的化学组成。方法采用酶解法和乙醇沉淀提取不同海参多糖,以总糖、硫酸根、糖醛酸、氨基糖和中性单糖含量为指标,进行化学组成分析比较,并在Q-Sepharose FF阴离子交换树脂色谱柱上分离,绘得洗脱曲线。结果10种海参多糖的总糖、硫酸根、糖醛酸和氨基糖含量的波动范围分别为40.33%~62.11%,19.54%~29.95%。9.85%~13.38%和4.92%~10.18%。中性单糖测定表明10种海参多糖均含有岩藻糖和半乳糖,部分含有甘露糖和葡萄糖,且岩藻糖含量很高。10种海参多糖在阴离子交换树脂色谱柱上的洗脱曲线明显不同,除仿刺参多糖和日本刺参多糖外,其余8种海参多糖均在盐浓度为3.2mol·L^-1的洗脱条件下洗脱完全。结论不同海参多糖在化学组成上差异明显,其在阴离子交换树脂色谱柱上分离的洗脱曲线也呈现不同特点。Objective To analyse the chemical component difference of the polysaccharides separated from ten kinds of sea cucumber, selected from the market. Methods Polysaccharides from different sea cucumbers were separated by papain , precipitated by ethanol, analysed with six chemical indexes,including content of total sugars, sulfate, uronic acid, aminosugars and neutral monosaccharide, and eluted on Q-Sepharose FF, recording the elution curve. Results Content of total sugars, sulfate, uronic acid and aminosugars of polysaccharides from ten kinds of sea cucumbers fluctuated within 40. 33%~62.11%, 19.54 %~29. 95%, 9.85%~13.38% and 4.92 %~10.18 %, respectively. Neutral monosaccharide analysis showed that fucose and galactose were in all the ten polysaccharides while mannose and glucose existed partly, and the content of fucose was very high. Elution curve of polysaccharides from ten kinds of sea cucumbers on Q-Sepharose FF differed obviously. All the polysaccharides were eluted at the salinity of 3.2 mol·L^-1 ,except for those from Apostichopus japonicus and Stichopus japonicus. Conclusion Chemical component of polysaccharides from different sea cucumbers showed obvious diversity, and the elution curves of these polysaccharides on Q-Sepharose FF were distinct.
