摘 要:目的:对比研究长期摄入高剂量不同富硒植物硒对大鼠重要肝指标的影响,为安全合理利用不同植物硒源提供科学依据。方法:采用N-甲基-N’-硝基-N-亚硝基胍(MNNG)诱发大鼠胃癌模型,连续灌喂四种高剂量不同富硒植物硒17w,于18w末测定大鼠肝脏硒累积、丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GPx)活性。结果:摄入富硒青花菜、富硒红羽、富硒绿羽的大鼠肝硒水平显著高于摄入富硒大蒜。补充植物硒显著降低了大鼠的肝MDA水平,提高了肝GPx活性。富硒大蒜高、低剂量组大鼠肝MDA含量差异显著,而GPx活性差异不显著。结论:长期摄入高剂量富硒大蒜后,动物肝组织低硒累积可作为富硒大蒜高安全性的重要依据。植物硒可通过GPx清除肝组织MDA,防止肝氧化损伤。Objective: To study the safety of selenium from application of selenium (Se)-enriched plants in rats with stomach cancer induced by M-methyl-N'-nitroso guanidine(MNNG). Methods-Seventy rats were fed the basal diet for a week, and divided equally into the control, MNNG and five Se treatment groups. Rats were given 15mg/kg bw of daily dose of MNNG for 10 d except the control group, and were given Se 150 or 300μg/kg bw of daily suspension of Se-enriched plants by gavage for 17 win five Se treatment groups. Rats were sacrificed at the end of 18th week, and the liver Se and malondialdehyde (MDA) contents as well as glutathione peroxidase (GPx) activities were determined. Results- The liver Se contents in rats supplemented by Se-enriched garlic were significantly lower than those fed with broccoli and two kales. The liver MDA levels of the Se supplemented groups were significantly lower than those in the MNNG and control groups, much lower with 300μg/g bw Se than 150μg/g bw while the liver GPx activities of the Se supplemented group were significantly higher without difference between Se doses. Conclusion: Low liver Se accumulation in rats fed with long-term and high-dose of Se-enriched garlic is an important evidence of low toxicity of Se-enriched garlic. Se from plants is effective in elimination of liver MDA by GPx.
