摘  要:在5L发酵罐中,采用高产壳聚糖酶菌株Penicillium sp.ZD-Z1,经半连续发酵和超滤耦合制备不同分子质量壳低聚糖。考察了稀释率、壳聚糖浓度、发酵时间对菌体生长和产量的影响。理想条件为:当稀释率为0.15h-1,流加壳聚糖浓度为3%的培养基,发酵96h后,壳低聚糖产量提高了5倍。采用不同截留分子质量(MWCO)超滤膜分离不同分子质量的壳低聚糖。通过喷雾干燥获得高纯度壳低聚糖样品。The coupling of semi-continuous fermentation and ultrafiltration was developed on chitosan-oligosaccharide preparation. High yielding chitosanases strain: Penicillium sp. ZD-Z1 was applied in the fermentation carried out in a 5-liter fermentor. The influences of dilution rate, chitosan concentration in broth and fermentation time on biomass and yield were studied. It was found that the optimal operation condition of fermentation was under dilution rate of 0.15 h^-1 , chitosan concentration in broth of 3% and fermentation time of 96 hours. The yield of chitosan-oligosaccharides was increased 5-fold after coupling. Separation of chitosan-oligosaccharides with different molecular weight was achieved based on the use of ultrafiltration mem- brane with different molecular weight cut-off (MWCO). Chitosan-oligosaccharide powders with high purity were acquired by spray drying.
