摘  要:用不同浓度的丙酮对小鼠肝脏蛋白进行分级提取沉淀,用SDS-聚丙烯酰胺梯度凝胶电泳实验寻找正常组和胶铬组的差异表达蛋白质,进而通过基质辅助激光解吸电离飞行时间质谱检测差异蛋白的肽质量指纹,对差异蛋白进行鉴定。通过无SDS-聚丙烯酰胺梯度凝胶电泳试验检测正常组和胶铬组肝脏的超氧化物歧化酶(SOD)活力,研究胶原蛋白多肽-铬(Ⅲ)螯合物对小鼠肝脏蛋白质表达的影响,探讨其抗氧化的机理,证明胶原蛋白多肽-铬(Ⅲ)螯合物可以显著提高小鼠肝脏内SOD的表达。This experiment research on the effects of collagen peptide-chromium (Ⅲ) complex (CPCC) on expression of protein. It deeply discusses the principle of antioxidation. Mice can be divided into two groups: CPCC group (Feeding with collagen peptide-chromium (Ⅲ) complex) and normal group (Feeding with same volume distilled water). We will extract protein in liver from different levels with acetone. Then we will try to find out the differences on the expression of protein between normal and CPCC group by SDS-PAGE grads electrophoresis experiment. Furthermore, with the help of Matrix assisted laser desorption/ionization time of flight (MALDI- TOF), mass spectrometry was used for the peptide mass fingerprinting of the different protein. And then by non SDS-PAGE grads electrophoresis, the vitality of Superoxide Dismutase (SOD) in liver form normal and CPCC group can be detected. Expression on SOD in liver of mice can be obviously improved by collagen peptidechromium (Ⅲ) complex.
