摘  要:开发了一种可同时检测四种转基因作物的高灵敏度非荧光标记(生物素标记)基因芯片方法。应用该方法检测了4种含一定浓度转基因产品(包括转基因大豆Roundup Ready、转基因玉米MON810、Bt11和转基因油菜RT73)的有证标准物质,并做了检测灵敏度试验。结果表明,该技术可同时有效检测这4种转基因产品中的11个目标基因,检测低限可达0.1%以下,在现有转基因产品检测方法(包括荧光标记基因芯片)中达到或超过最低水平,但其检测成本远低于荧光标记基因芯片法。A highly sensitive non-fluorescence (biotin-marking) gene-chip method was developed to detect 4 genetically modified organisms (GMO) simultaneously. 4 certificated reference materials (CRM), which contain certain portion of specific GMOs (including genetically modified soybean Roundup Ready, genetically modified corn MON810 and Bt11, genetically modified rapeseed RT73), were detected using this method, while the sensitive testing was done. Results demonstrated that by this technology 11 target genes in 4 GMO products were able to be effectively detected simultaneously, and the limit of detection was less than 0.1%, which shows that this method is currently one of the most sensitive ways to detect of GMOs, nevertheless the cost is far lower than fluorescence gene-chip technology.
