摘 要:目的:建立杜仲茶中桃叶珊瑚甙含量测定的超高压提取(UHPE)-高效液相色谱(HPLC)分析法。方法:采用正交试验考察超高压提取压力、提取时间、料液比和甲醇浓度对提取桃叶珊瑚甙的影响,以HPLC法为含量测定方法。结果:最佳提取桃叶珊瑚甙的条件为超高压提取压力350MPa、时间3min、料液比1:8、70%甲醇。HPLC法测定桃叶珊瑚甙的线性范围在5~100μg/ml,相关系数为0.9998。杜仲茶中桃叶珊瑚甙的含量为1.94%,相对标准偏差为3.02%,加标回收率为97.40%~100.16%。结论:此法简单、快速、经济,是测定杜仲茶中桃叶珊瑚甙含量的有效方法。Objective:To establish a sensitive analytical method for detection of the aucubin in eucommia tea by using ultra high pressure extract(UHPE) and high performance liquid chromatograpy(HPLC). Methods :Orthogonal test was used to evaluate the influence of the 4 factors including the UHPE pressure, UHPE time, the ratio of solid to liquid and the extracting concentration of methanol. HPLC method was used to determine the aucubin in eucommia tea. Results:The optimum extraction was as follows: UHPE pressure 350 MPa, UHPE time 3 min, stock ratio 1:8 (g:ml) and 70% methanol. The linearity was superior from 5- 100 μg/ml. In eucommia tea, the content of aucubin was about 1.94% , the standard deviation was 3.02%. The average recovery rate was 97.40%-100.16%. Condution:This method is quick and economical. It has been satisfactorily applied to the aucubin analyses.
