摘 要:在摇瓶及5L发酵罐中研究了补加葡萄糖对始旋链霉菌(Streptomyces pristinaespiralis)XC416发酵生产普那霉素的影响。摇瓶发酵结果表明,初始葡萄糖浓度以40g/L为宜,通过在对数生长期补加20g/L的葡萄糖可以使生物量增加23%~31%,普那霉素的产量可以提高20%~36%;在稳定期补加20g/L的葡萄糖虽然对生物量影响不大,但能显著提高菌体合成抗生素的能力,使普那霉素的产量提高56%~76%;而在对数生长期中后期和稳定期前期分两次各补加10g/L的葡萄糖,则能使普那霉素的产量提高1.92倍。5L发酵罐补料分批发酵结果表明,在对数生长期和抗生素合成的初期分三次共流加36g/L葡萄糖不仅有效地延长了抗生素的合成期,还提高了菌体合成抗生素的能力及产物得率,最终使普那霉素的产量提高1.02倍。The influences of glucose feeding on pristinamycins fermentation process of Streptomyces pristinaespiralis XC 416 in shake flasks and 5L bioreactor were investigated. In shake flasks, the initial glucose concentration of 40 g/L was optimal for pristinamycins fermentation. When 20 g/L glucose was fed at exponential phase of the producing strain, a 23%~ 31% higher biomass could be obtained and the yield of pristinamycins was increased by 20% ~36% ; when 20g/L glucose fed at stationary phase, even no obvious effect on the biomass was observed, the ability of the mycelia producing antibiotics could be remarkably improved and ultimately the yield of pristinamycins was increased by 56% ~ 76% ; when 10g/L glucose was fed at the later period of exponential phase and the early period of stationary phase, the final yield was 1.92 times higher than that without glucose feeding; when 36g/L glucose was fed into a 5L bioreactor three times during exponential phase and the early period of production phase, not only the production time effectively prolonged, but also the ability of the mycelia producing antibiotics with glucose were improved and the production of pristinamycins were 1.02 times higher than control.
