摘 要:采用正己烷、氯仿、乙酸乙酯、丙酮、甲醇5种溶剂分别提取银杏内生炭角菌YX-28的子座粉末,测定各提取物中的总酚和总黄酮含量,并通过二苯基苦基苯肼(DPPH)和β-胡萝卜素/亚油酸模型评估其抗氧化活性,以期获得一种新型、高效、安全的抗氧化剂来源。结果表明:提取物中总酚和总黄酮含量均与抗氧化活性存在正相关关系,甲醇提取物具有最高的总酚和总黄酮含量,分别为(54.51±1.05)mg.g-1(没食子酸当量)和(86.76±0.58)mg.g-1(芦丁当量)。在β-胡萝卜素/亚油酸试验中,400μg.mL-1的YX-28甲醇粗提物具有72.90%的抑制活性,显著高于阳性对照二丁基羟基甲苯(BHT)和抗坏血酸(AsA);而在DPPH自由基清除试验中则显示中等活性。通过气相色谱-质谱联用法(GC/MS)分析鉴定了甲醇提取物中的41种物质,其占总提取物的64.65%。Different solvents of hexane, chloroform, ethyl acetate, acetone and methanol were used to extract the stromata of Xylaria sp. YX-28, an endophytic fungus isolated from Ginkgo biloba. Total phenolic and flavonoid contents were determined, and antioxidant activity was evaluated. The results showed that positive correlations were found between total phenolic and flavonoid contents in the extracts and their antioxidant activities. The highest phenolic and flavonoid contents existed in the methanol extract, (54. 51 ± 1.05 ) mg · g^-1 ( galic acid equivalent) and ( 86. 76 ± 0. 58 ) mg · g^-1 ( rutin equivalent), respectively. In the β- carotene/linoleic acid assay, the methanol extract showed 72.90% inhibition at the concentration of 400 μg·mL^-1, higher than butylated hydroxytoluene (69.77%) and ascorbic acid ( 14. 85% ) standards, while 2,2-diphenyl-1-picrylhydrazyl free radical-scavenging activity of the methanol extract was moderate, compared with positive controls. GC/MS analysis of the methanol extract resulted in the identification of 41 compounds, representing 64. 65% of the extract.
