摘 要:选取提取温度、时间、料液比和NaOH浓度作为考察因子,在单因素实验的基础上,进行了均匀设计实验,采用逐步回归方法建立了具有良好预测性能的碱浸提工艺条件的数学模型,通过Matlab7.0中的fmincon软件确定了海参糖胺聚糖最佳的提取工艺条件。MTT比色法考察了糖胺聚糖对黑色素瘤B16细胞抑制作用的生物活性。实验结果表明:提取温度对海参糖胺聚糖得率的影响显著,最优提取工艺条件为:提取温度85℃,提取时间10min,料液比1∶55(g∶mL),NaOH浓度10%,在该最佳工艺条件下进行验证实验,糖胺聚糖的得率可达36.08%,与模型的预测值36.01%基本一致,表明模型可较好地预测海参糖胺聚糖的得率。糖胺聚糖对黑色素瘤细胞作用72h,IC50为0.13mg/mL,具有显著的抗肿瘤活性。The extracting temperature, extracting time, the ratio of water to solid, concentration of NaOH were chosen as the observed factors. Base on one- factor tests, uniform experiments design method was applied to design the experiments. Step regression method was used for establishing mathematic model which had good predictability for extracting conditions with alkali. The optimum extraction conditions of holothurian glycosaminoglycan were confirmed by Matlab 7.0 fmincon software. The effects of glycosaminoglycan on the growth of Melanoma B16 were observed by MTT. The results showed that extraction temperature impacted significantly the rate of holothurian glycosaminoglycan. The optimum extraction conditions were confirmed as follows: extraction temperature 85℃, extraction time 10min, the ratio of solid to water 1 : 55 ( g : mL), NaOH concentration of 10%. The extraction rate of glycosaminoglycan under the optimum conditions could reach the highest value of 36.08% ,consistenting with the predictive value 36.01%. So the model could predict accurately the rate of holothurian glycosaminoglycan. Glycosaminoglycan had significant anti-tumor activity on Melanoma B16 after 72h and the IC50 was 0.13mg/mL.
