摘 要:以青霉TS406Y(Penicillum sp.)为诱变出发菌株,评价了硫酸二乙酯(DES)、亚硝基胍(NTG)和甲基磺酸乙酯(EMS)对该菌株的诱变效应,发现在常规剂量内达到70%致死率时所需要的时间分别为23,3,90 min.根据使用方便和安全原则,进行了青霉TS406Y的DES诱变和选育,获得了胆固醇氧化酶活性提高84.5%的突变菌株D5,其胆固醇氧化酶活性达到了1240 U/L,连续传代6次,发现D5菌株具有遗传稳定性.另外还发现,利用紫外线(UV)和EDS对单因素诱变后获得的胆固醇氧化酶高产菌株进行复合诱变,94%的突变菌株为负变菌株.Mutated effect was determined of mutagenic agent which were diethyl sulfate, N-Methyl-N-ni- troso-N'-nitroguanidine and Methanesulfonic acid ethylester, when the Penicillum sp. TS406Y was treated for 23,3,90 min by above mutagenic agent respectively, 70 % lethal rate was gotten. Cholesterol-degrading mutant D5 was obtained from Penicillum sp. TS406Y by treatment with diethyl sulfate. The activity of cholesterol oxidase of strain DS, which was 1 240 U/L, and was 84.5 % higher than TS406Y. Meanwhile the results showed that 94 % negative mutants were obtained by the combined treatment with ultraviolet and diethyl sulfate.
