摘  要:[目的]研究亚硝酸钠(NaNO2)致中国仓鼠肺细胞(CHL)染色体畸变的作用。[方法]测定NaNO2对CHL细胞的半数抑制浓度(IC50),根据IC50设立不同剂量组,进行正式实验,分别观察不加及加S9后的染色体畸变情况,根据标准进行结果判定。[结果]NaNO2染毒在不加S9时,128、12.8和1.28μg/ml3个剂量组的染色体畸变率均﹥5%而≤10%,为可疑阳性,而高剂量组(1.28mg/ml)畸变率﹥10%而﹤20%,为阳性反应。NaNO2染毒在加入活化系统S9后,各剂量组引起CHL细胞染色体畸变率在12.8和1.28μg/ml2个剂量组的染色体畸变率均﹥5%而≤10%,为可疑阳性,而中(128μg/ml)和高(1.28mg/ml)剂量组畸变率均﹥10%而﹤20%,为阳性反应。[结论]NaNO2在1.28mg/ml剂量下可引起CHL细胞染色体畸变率增高。[ Objective ] To explore the effects of Sodium Nitrite (NANO2) on chromosome aberration in lung ceils of Chi, nese hamster. [Methods] The effect of NANO2 on the IC50 of CHL Ceils was determined, these Chinese hamsters were divided into different groups in line with different dosage to perform the official test so that to observe the chromosome aberration status with S9 and without, and the results were determined according to the standardized criteria. [Results] When the concentrations of NaNO, were 128, 12.8 and 1.28 μg/ml without S9, the chromosome aberration rates were at the range of 5% to 10%, which was suspected to be positive. While the Chinese hamsters in group with high concentration of 1.28mg/ml, the chromosome aberration rate was at the range of 10% to 20%, which showed to be positive reaction. When adding S9 after treating with NANO2, the chromosome aberration rates of CHL cells induced by the dosage groups with the concentration of 12.8 μg/ml and 1.28 μg/ ml were at the range of 5% to 10%, which was suspected to be positive. While the chromosome aberration rates in groups at the concentration of 128 μg/ml and 1.28mg/ml were at the range of 10% to 20%, which showed to be positive reaction. [Conclusion ] NaNO2 can induce chromosome aberration in CHL cells at the concentration of 1.28mg/ml.
